What is the quickest and easiest way to obtain a SAM file. Physically and if its possible over a wireless  connection? What is the best way to protect these files?

Without a doubt the fastest way is to have physical access to the computer and use a live CD to boot to. And yes you can extract it over a wireless connection but thats more involved and will require exploiting the box.
